Structures of these compounds were elucidated based on their spectral properties. The essential oil from the fruits of the endemic species Ferula drudeana collected from Aksaray was analyzed by GC and GC/MS. The main components of the oil were determined as shyobunone (44.2%) and 6-epishyobunone (12.6%). The essential oil of the fruits and various solvent extracts of the fruits and roots of F. drudeana were evaluated for their antibacterial and anticandidal activity using microbroth dilution methods. The essential oil of the fruits, methanol, and methylene chloride extracts of the fruits and roots showed weak to moderate inhibitory activity against all tested microorganisms with MIC values of 78–2000 µg/mL. However, the petroleum ether extract of the roots showed remarkable inhibitory activity against Candida krusei and Candida utilis with MIC values of 19.5 and 9.75 µg/mL, respectively. Furthermore, all the samples were tested for their antioxidant activities using DPPH• TLC spot testing, online HPLC–ABTS screening, and DPPH/ABTS radical scavenging activity assessment assays. Methanolic extracts of the fruits and roots showed strong antioxidant activity in both systems.Öğe Biological activities of the natural coumarins from apiaceae plants(ACG Publications, 2023) Tosun, Fatma; Biltekin, Sevde Nur; Karadağ, Ayşe Esra; Mıhoğlugil, Feyyaz; Akalgan, Demet; Miski, MahmutNineteen natural coumarins; badrakemin (1), colladonin (2), 14'-acetoxybadrakemin (3), anatolicin (4), 14'-hydroxycolladonin (5), badrakemone (6), karatavicinol (7), 14'-acetoxybadrakemone (8), 14'-acetoxycolladonin (9), colladonin acetate (10), deltoin (11), smyrnioridin (12), isoimperatorin (13), oxypeucedanin (14), bergapten (15), osthol (16), 4'-senecioyloxyostol (17), neopapillarine (18), and scoparone (19) were tested against U87MG, A549, and PC3 cancer cell lines as well as against healthy human embryonic kidney cell line, HEK293. Colladonin (2) was found to have IC50 values of 12.6 and 11.58 µM in A549 and PC3 cell lines, respectively. Deltoin (11) and 14'-acetoxybadrakemin (3) were found to have an IC50 value of 9.92 µM and 11.85 µM against the U87MG cell line, respectively. Remarkably, these compounds show very low cytotoxicity against the healthy human embryonic kidney cell line, HEK293. In addition to the cytotoxic activity, nineteen natural coumarins were tested for their inhibitory activity against 5-LOX, collagenase, and elastase enzymes.Öğe Biological activity and chemical composition of the essential oil from the fruits of Ferula halophila Peşmen(Istanbul Medipol University, 2023) Kürkçüoğlu, Mine; İşcan, Gökalp; Tosun, Fatma; Miski, MahmutThe hydrodistilled essential oils of the dried fruits of F. halophila Peşmen, an endemic species growing near Salt Lake in Central Anatolia, were analyzed by GC and GC-MS systems; 27 and 24 compounds representing 92.1% and 79.8% of the essential oils were characterized, respectively. The main component of the essential oils was identified as ?-phellandrene. The antibacterial and anticandidal effects of the essential oils were determined by using partly modified CLSI methods M7-A7 and M27-A2, respectively. The essential oils from two specimens showed weak to moderate inhibitory effects on the tested pathogenic bacteria (MIC, 125-2000 µg/ mL) and Candida panels (MIC, 156-1250 µg/mL).Öğe Biological activity and chemical composition of the essential oil from the fruits of Ferula rigidula Fisch. ex DC.(Istanbul Medipol University, 2023) İşcan, Gökalp; Kürkçüoğlu, Mine; Tosun, Fatma; Miski, MahmutThe hydro-distilled essential oils of the fruits of F. rigidula Fisch. ex DC. was analyzed by Gas Chromatography (GC) and Gas Chromatography/Mass Spectrometry (GC/MS) systems at the same time. Thirty-one compounds were characterized rep-resenting 98.2% of the essential oils. The main components of the oil were determined as a-pinene and camphene (24% and 20%, respectively). The anticandidal and antibacterial effects of the essential oil were determined by using partly modi-fied CLSI protocols M27-A2 and M7-A7, respectively. The essential oil of the dried fruits showed several inhibitory effects on the tested bacteria panel (MIC, 62.5 to 2000 µg/mL) and Candida species (MIC, 125 to 1000 µg/mL).Öğe Butyrylcholinesterase-inhibiting natural coumarin molecules as potential leads(Elsevier Ltd, 2021) Erdoğan Orhan, İlkay; Tosun, Fatma; Şenol Deniz, Fatma Sezer; Eren, Gökçen; Mıhoğlugil, Feyyaz; Akalgan, Demet; Miski, MahmutSeventeen natural coumarin derivatives; badrakemin (1), 14?-acetoxybadrakemin (2), badrakemone (3), 14?-acetoxybadrakemone (4), colladonin (5), colladonin acetate (6), 14?-acetoxycolladonin (7), karatavicinol (8), deltoin (9), smyrnioridin (10), marmesin (11), osthol (12), oxypeucedanin (13), oxypeucedanin hydrate (14), isoimperatorin (15), scopoletin (16), and umbelliprenin (17), were tested against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), the sister enzymes that play a critical role in the pathology of Alzheimer's disease as well as tyrosinase (TYR) as the target for Parkinson's disease. The tested coumarins were more selective against BChE, where the coumarins 2, 5, 8, and 15 (IC50 = 30.3 ?M, 29.2 ?M, 37.2 ?M, and 50.1 ?M, respectively) displayed higher BChE inhibition than the reference (galanthamine, IC50 = 60.2 ?M) at 100 ?g/mL. Only four coumarins (2, 5, 9, and 15) showed inhibition against AChE. Binding conformations of the coumarins (2, 5, 8, 9, and 15) within the active sites of AChE and BChE were explored via molecular docking experiments. The docked compounds were oriented by the interactions with the oxyanion hole and the peripheral anionic site residues of AChE/BChE. The coumarin derivatives 1–17 was found to have no or low inhibition (2.03 ± 0.92 %–12.91 ± 0.40 %) against TYR at 100 ?g/mL. Our findings revealed that coumarins could be promising lead compounds for designing novel anti-Alzheimer drug candidates.Öğe Chemical characterization of Glaucosciadium cordifolium (Boiss.) B. L. B. L. Burtt& P. H. Davis (Apiaceae) were analyzed using gas chromatography-flame ionization detector/mass spectrometry, simultaneously. Furthermore, antimicrobial and antioxidant activities of G. cordifolium volatile oils were investigated for possible utilization. Total of 62 volatile compounds were identified in G. cordifolium essential oils, where the main component was characterized as alpha-pinene in all parts, commonly. The other main components were beta-pinene (15.7%), (Z)-beta-ocimene (14%) and sabinene (7%) in the volatile oil of the aerial part; sabinene (10.1%), beta-pinene (10.1%) and alpha-phellandrene (5.3%) in the essential oil of the fruits; hexadecane (12.2%), tetradecane (11.9%), octadecane (7.4%) in the essential oil obtained from the root, respectively. The in vitro microdilution method was used for the antimicrobial activity testing against Salmonella typhi ATCC 6539, Acinetobacter baumanii ATCC 19606, Bacillus cereus ATCC 14579, Staphylococcus aereus ATCC 6538, Listeria monocytogenes ATCC 19115, Helicobacter pylori ATCC 43504 and Mycobacterium avium ATCC 25291. The best antimicrobial activity of the volatile oils was against L. monocytogenes among the tested microorganisms. In addition, DPPH center dot-ABTS(center dot) scavenging activity was tested, none of the essential oils showed any significant antioxidant activity.Öğe Chemical compositon, antibacterial and antioxidant activities of Cnidium silaifolium ssp. orientale (Boiss.) Tutin were analyzed both by GC-FID and GC/MS, simultaneously. One hundred nine compounds representing 90.1% of the total volatiles in the EOs were identified with the main characteristic compounds alpha-pinene (50.3%) in the root, germacrene D (20.3%) in the fruit, and beta-caryophyllene (18.7%) in the aerial parts of C. silaifolium ssp. orientale. The antimicrobial activity against human pathogenic Gram-negative and Gram-positive bacteria was evaluated by the in vitro microdilution method. Antibacterial succeptibility was observed from the root and aerial part EOs against Staphylococcus aureus (0.039 and 0.156 mg/mL, respectively); while the fruit EO was most effective against Bacillus cereus at 0.07 mg/mL. The highest cytotoxic activities were observed in the dichloromethane extract of the fruits with IC50 values of 12.1 mg/mL and 12.2 mg/mL on the COLO205 and KM12 cell lines, respectively. In contrast, the ethyl acetate extract of the fruits exhibited moderate cytotoxic activity with IC50 values of 33.6 mg/mL and 37.2 mg/mL against the COLO205 and KM12 cell lines. Dichloromethane and ethyl acetate extracts of the fruits exhibited weak cytotoxic activity against the UO31 and A498 cancer cell lines with IC50 values greater than 50 mg/mL. Aqueous-methanol extract of the fruits showed cytotoxic activity against all cancer cell lines with IC50 value greater than 50 mg/mL.Öğe Cytotoxic activity of the root and fruit extracts of Heptaptera anisoptera (DC.) The dichloromethane extract of the roots of H. anisoptera showed cytotoxic activity with IC50 values of 3.1 and 3.9 ug/mL on the COLO205 and KM12 cell lines, respectively. Furthermore, revised spectroscopic data for 14'-acetoxybadrakemone (4) and complete NMR spectroscopic assignments for colladonin (6) and colladin (7) are reported.Öğe Cytotoxicity screening of some Turkish plants against renal cancer cells(Marmara University, 2023) Mihoğlugil, Feyyaz; Akalgan, Demet; Tosun, FatmaIn this contribution, we performed cytotoxicity screening of 51 extracts from 17 plant species, representing 14 families growing in Turkey. Cytotoxic activities of the dichloromethane, ethyl acetate, and methanol extracts of the plants were investigated against the renal (A498 and U031) cancer cell lines. 28 extracts exhibited more than 50% growth inhibition at a 25 ug/mL concentration on both renal cancer A498 and UO31 cell lines. The extracts showed higher activity against UO31 cells than A498 cells. The dichloromethane extract of the aerial parts of Scabiosa columbaria ssp. columbaria showed the highest cytotoxic activity with 64% inhibition at a 25 ug/mL concentration on the renal cancer UO31 cell line. The methanol extract of the aerial parts of Epilobium minutiflorum showed the highest cytotoxic activity with 63% inhibition at a 25 ug/mL concentration on the renal cancer A498 cell line.Öğe Determination of the volatile compounds of anthemis cretica subsp. The essential oil of A. cretica subsp. anatolica was characterized by the presence of a high percentage of oxygenated sesquiterpenes (57.9%). Twenty-seven compounds were identified representing 96.6 % of the essential oil of A. cretica subsp. anatolica. The air-dried aerial parts were extracted with methanol. Gels were prepared, followed by the hemostatic activity evaluation using Wistar albino rats in vivo tail tip amputation model. The chemical characterization of the total extract was performed by high-performance thin-layer chromatography. The methanol extract was loaded to a hydrogel base to complete a concentration of 2.5 and 7.5%. As a result, the 2.5% extract gel was found more effective on both bleeding time and amount. Hyperoside and tiliroside, which may be the components responsible for the hemostatic properties of the analyzed plant material, were dereplicated as major constituents.Öğe In vitro anti-helicobacter pylori and antimycobacterial activity evaluation of selected plants from Turkey(2020) Karadağ, Ayşe Esra; Çaşkurlu, Ayşegül; Tosun, FatmaIn this study, the extracts of Ulmus minor Mill. subsp. minor (Ulmaceae), Lathyrus pratensis L. (Leguminosae), Glaucium leiocarpum Bois. (Papaveraceae), and Echium vulgare L. (Boraginacea) were investigated for their in vitro anti-Helicobacter pylori and antimycobacterial activity. The air-dried plant material was powdered and extracted with 70% ethanol by maceration. The extract was filtered and evaporated to dryness under the vacuum and then dissolved in a waterethanol (90:10) mixture and extracted with dichloromethane and ethyl acetate, respectively. Antibacterial activity was investigated by microdilution method against Helicobacter pylori ATCC 43504, Mycolicibacterium smegmatis ATCC 14468 and Mycobacterium avium ATCC 25291. The ethyl acetate extract of U. minor showed activity with MIC of 250 µg / mL against H. pylori. The other extracts showed no or weak inhibitory activity at >2000 µg/mL concentrations against the tested microorganisms. In conclusion, ethyl acetate extract of U. minor may be used for the treatment of H. pylori infections.Öğe In vitro antifungal activity of cistus creticus l. Against plant pathogenic fungi(Istanbul Medipol University, 2020) Çaşkurlu, Ayşegül; Karadağ, Ayşe Esra; Köse, Yavuz Bülent; Tosun, FatmaCistus creticus L. is a well-known member of the Cistaceae family. In this present study, methanol extract of aerial parts of C. creticus was evaluated for its antifungal activity against plant pathogenic fungi such as Fusarium moniliforme NRRL 2374, Fusarium culmorum NRRL 3288, Alternaria alternata ATCC 6663, and Botry-tis cinerea AHU 9424, respectively. The in vitro antifungal activity of C. creticus extract was determined by calculating mycelial growth inhibition. It was demonstrated that methanol extract of C. creticus has shown antifungal activity on F. mon-iliforme (36,7%) and B. cinerea (30,2%). Moreover, we are planning to evaluate the active fractions which are responsible for the antifungal activity in further studies.Öğe In vitro antimicrobial and antioxidant activity evaluation of melampyrum arvense L. Var. elatius boiss. and sedum spurium m. bieb. extracts(İstanbul Medipol University, 2019) Karadağ, Ayşe Esra; Tosun, FatmaSedum spurium M. Bieb. (Crassulaceae) is a common ornamental plant, whereas, Melampyrum arvense L. var. elatius Boiss. (Orobanchaceae) is a semi-parasitic plant and grows naturally in the fields. In this study, the dichloromethane and ethyl acetate extracts of M. arvense and S. spurium were evaluated for their in vitro antioxidant and antimicrobial activities. The antioxidant activity was evaluated by DPPH•-ABTS• methods. The antimicrobial activity of S. spurium and M. arvense extracts was determined using the in vitro broth microdilution assay against following human pathogenic strains; Staphylococcus aureus ATCC 6538, Enterococcus faecalis ATCC 29212, Escherichia coli NRLL B-3008, Helicobacter pylori ATCC 43504, Mycobacterium smegmatis ATCC 25291, Mycobacterium avium ssp. avium and Pseudomonas aeruginosa ATCC 10145. The extracts showed weak antimicrobial activity against Gram-negative/positive bacteria, having the MIC values of 500-1000 ?g/mL. Antibacterial activity was not observed against Mycobacteria at 2000 µg/mL. In addition, antioxidant activity of M. arvense ethyl acetate extract was higher than those of the other extracts.
