Trigonelline's effect on BCL-2 protein expression level in THP-1 cells

Objective: Trigonelline (TRG), is one of the main constituents of coffee and is an alkaloid derivative of vitamin B6 in plants. TRG has been reported to have antihyperglycemic, antihyperlipidemic, and antioxidant activities. At the same time studies of TRG on cancer and the effect on diabetes have been done. However, there is no study of the effect of TRG on apoptosis in monocytes. In this study, the possible effects of trigonelline on apoptosis on human monocytes were examined. Seven experimental groups were evaluated: (1) the control group treated with the medium (RPMI-1640), (2) the cell group treated with 200 ?M H2O2 for 1 hour, (3/4/5/6/7) The cell group treated with 10?5/5 × 10?5/10?4/2,5 × 10?4/5 × 10?4 M TRG for 24 hours and then treated with 200 ?M H2O2 for 1 hour (anti- Apoptotic effects). Bcl-2 protein expression levels were measured by western blot. It was observed that TRG significantly reduced Bcl-2 protein expression levels increased by H2O2 (P < 0.005) at all doses given. The results of preliminary experiments reveal the effects of trigonelline, which is involved in autophagic and apoptotic processes. It has been indicated that trigonelline may have a potential role in the prevention and treatment of oxidative stress in monocytes.
