Öğe Effects of melatonin on both testicular regeneration and recovery of spermatogenesis in busulfan-treated rats(Faculdade de Ciencias Farmaceuticas (Biblioteca), 2022) Taşlıdere, Elif; Eşrefoğlu, ?Mukaddes; Tok, Olgu Enis; Taşlıdere, Bahadır; Bulut, HuriTesticular damage is one of the most hazardous effects of chemotherapy as it is frequently associated with oligozoospermia and azoospermia. This study aimed at evaluating the protective effect of melatonin in a rat model of busulfan-induced testicular injury. Rats were divided into four groups: control, melatonin, busulfan, busulfan plus melatonin. After 15 days, the semen was collected from the epididymis and testes were assessed. Sperm removed from cauda epididymis and analyzed for sperm count and viability. Testis tissues were also removed, fixed in formalin and were embedded in paraffin. Sections of testis tissue were stained with hematoxylin-eosin for histological examination and prepared for TUNEL (Terminal deoxynucleotide transferase dUTP Nick End Labeling) assay to detect apoptosis and PCNA (proliferating cell nuclear antigenassay) to detect proliferation cells. Serum and testes supernatants were separated to detect testosteron level and oxidative stress parameters. In histological examination, degenerative changes in seminiferous tubules were observed in the experimental groups. In biochemical examination, the total oxidant status (TOS) levels in Busulfan group were significantly higher than in the control group while the total antioxidant status (TAS) levels of all the groups were similar. In conclusion, the beneficial properties of melatonin treatment by its potent anti-oxidants may reduce adverse effects of chemotherapy in the reproductive system in a rodent system.Öğe Evaluation of the association of mucosa-associated invariant t (mait) cells with childhood asthma(2024) Ekşi, Meral; Bulut, Huri; Akalın, Erdem; Ustabaş Kahraman, Feyza; Yazan, Hakan; Yazıcı, Mebrure; Çakır, ErkanBackground/aim: Innate-like T lymphocytes are a recently defined group of T cells comprising mainly mucosa-associated invariant T (MAIT) cells. The relationship between MAIT cells and childhood asthma is controversial. In this study, we aimed to determine the role of MAIT cells in patients with allergic asthma (AA) and nonallergic asthma (NAA). This is the first study to compare the ratios of these cells in patients with AA and NAA. Materials and methods: The study included children aged 6–18 years with AA (n = 41) or NAA (n = 30) and healthy control subjects (n = 36). The control group consisted of children who presented to the outpatient clinic without chronic disease, malnutrition, or acute or chronic infection. The proportions of MAIT, TH17, MAIT-17, and Th17-17 cells were investigated by flow cytometry and compared among the AA, NAA, and control groups. Results: When the AA and NAA patient groups were compared, the mean MAIT cell ratio was significantly lower in NAA patients (median: 0.45, p < 0.05). MAIT cell ratios were also substantially lower in NAA patients compared to the control group (mean: 0.504, p < 0.05). TH17, MAIT-17, and TH17-17 cell values were not statistically significant among the groups. Conclusion: Our study found that MAIT cell ratios were lower in the NAA patient group compared to the control group and AA patients. It has been predicted that MAIT cell depletion may have a role in the development of NAA. Our study is the first on this subject in the literature and further studies are needed.Öğe Quercus pyrenaica honeydew honey with high phenolic contents cause dna damage, apoptosis, and cell death through generation of reactive oxygen species in gastric adenocarcinoma cells(SAGE, 2019) Koçyiğit, Abdurrahman; Aydoğdu, Gökhan; Balkan, Ezgi; Yenigün, Vildan Betül; Güler, Eray Metin; Bulut, Huri; Koktaşoğlu, Fatmanur; Gören, Ahmet Ceyhan; Atayoğlu, Ali TimuçinMany studies have shown that honey with high phenolic contents prevents cancer formation. Furthermore, recent studies have demonstrated that honey can be used for the treatment of cancer as well as cancer prevention. Antineoplastic effects of honey are often associated with their antioxidant phenolic contents. However, very few studies have dealt with the association of phenolic contents of honeys in terms of antiproliferative effects. The aim of this study was, therefore, to elucidate the cytotoxic, genotoxic, apoptotic, and reactive oxygen species (ROS) generating effects of honey samples on the basis of their phenolic and flavonoid contents. Fourteen different honey varieties were collected from various parts of Turkey, and their characteristics regarding total phenols, flavonoids, and antioxidant contents were determined to test their effects on gastric cancer cells (AGS). For convenience, 2 honey varieties were selected, namely, Ida Mountains Quercus pyrenaica honeydew honey (QPHH-IM) having the highest phenolic and antioxidant content and Canakkale multifloral honey (MFH-C) with the lowest phenolic and antioxidant content. Levels of 11 different phenolic compounds in QPHH-IM and MFH-C samples were determined by LC-MS/MS. AGS cells were incubated with different concentrations of QPHH-IM and MFH-C for 24 hours, then the cell viability, DNA damage, apoptosis, and generation of ROS were determined. We found that QPHH-IM had more cytotoxic, genotoxic, and apoptotic effects than that of MFH-C. We think that these effects are probably related to pro-oxidant activities due to the high phenolic contents present. Therefore, further research on high-phenolic honey may contribute to the future development of cancer therapeutics.Öğe Role of urotensin-2 in 5-fluorouracil-related arterial vasoconstriction in cancer patients(Karger, 2018) Şeker, Mesut; İsen, Hayati Can; Çevirme, Nidal; Aydın, Sinem; Bilici, Ahmet; Bulut, Huri; Yasin, Ayşe İrem; Çoban, Ezgi; Demir, Tarık; Aliyev, Altay; Koçyiğit, Abdurrahim; Türk, Hacı MehmetBackground: The aim of this study was to identify the possible relationship of 5-fluorouracil (5-FU)-related arterial vasoconstriction with urotensin-2 (UT-2), which has a high potential as an endogenic vasoconstrictor. Methods: We assigned the patients to 1 of 3 groups. Patients in group 1 received a bolus of 5-FU, those in group2a continuous infusion (CI) of 5-FU, and those in group 3 no 5-FU, which was also a control group. Pre- and post-treatment UT-2 levels and brachial arterial diameters were measured and recorded in all patients. Results: 132 patients were included in the study. Pre-and post-treatment brachial artery diameters were similar in all groups: in group 1 (3.28 +/- 0.52 vs. 3.25 +/- 0.44 mm, p = 0.740), in group 2 (3.57 +/- 0.47 vs. 3.46 +/- 0.45 mm, p = 0.441) and in the control group (3.51 +/- 0.52 vs. 3.25 +/- 0.44 mm, p = 0.818). Pre-and post-treatment UT-2 levels were significantly different in each group: in group 1 (39.5 +/- 30.9 vs. 56.7 +/- 27.1 ng/ml, p = 0.0001), in group 2 (37.7 +/- 33.7 vs. 62.5 +/- 37.7 ng/ml, p = 0.0001) and in the control group (52.9 +/- 40.2 vs. 60.8 +/- 40.7 ng/ml, p = 0.006). Conclusion: Our findings suggest that UT-2 has a high potential as a vasoconstrictor agent in our bodies and its level increases through a bolus or CI 5-FU. Methods: Between September 2017 and September 2019, the study recruited 54 patients with endometrial cancer. Measurements were taken of blood levels of human leptin, perilipin-1, adiponectin, adipolin, resistin, visfatin, and estrone preoperatively and postoperatively before adjuvant therapy or at the end of one month. The serum samples were separated by centrifugation for 10 mins at 3,000 revolutions/min, then stored at ?80 °C until assay. Results: In this prospective study, a total of 54 endometrial cancer patients were analyzed in two separate groups according to the omentectomy status. Comprehensive staging surgery with omentectomy and without omentectomy was performed in 26 patients and 28 patients, respectively. The age, body mass index, body fat index, waist circumference, and skin thickness values of the patients with and without omentectomy were found to be similar. No statistically significant difference was determined between the patients with and without omentectomy in respect of the blood level of the adipokines measured preoperatively. A strong statistically significant correlation was determined between the pre and postoperative levels of Human Leptin (p = 0.002), perilipin-1(p = 0.001), adipolin (p < 0.001), adiponectin (p < 0.001), resistin (p = 0.001), visfatin (p < 0.001), and estrone (p = 0.004) (r = ?0.43, ?0.47, 0.75, 0.84, ?0.47, - 0.58, ?0.41, respectively) Conclusions: Omentectomy affected the postoperative blood levels of adipokines in obese patients with endometrial cancer. As omentectomy may have some positive effects on metabolism in these patients, it may be considered during endometrial cancer surgery due to the possible positive metabolic effects.
