Obezite, hiperlipidemi, Tip-2 DM, hipertansiyon ve koroner kalp hastalıklarına sebep olan kronik bir hastalıktır ve son yıllarda dünyada ve ülkemizde hasta sayısı hızlı bir şekilde artmaktadır. Obezite, beraberinde ekonomik problemleri de getirmektedir. Tüm bu nedenlerden dolayı araştırmacılar, obezitede farmakoterapiye odaklanmıştır. Güncel olarak Amerikan Besin ve İlaç Dairesi'nin onayladığı, Orlistat, Lorcaserin, Fentermin/Topiramat isimli obezite ilaçları mevcuttur. Ancak bu ilaçların kardiyovasküler ve gastrointestinal yan etkileri olduğu bildirilmiştir. Bu araştırmada, GOAT (Ghrelin O-Açil Transferaz) enziminin obezite üzerine etkisi araştırılmıştır. GOAT enzimini inhibe edecek kimyasal molekül sentezlenmesi, moleküllerin bilgisayar ortamında moleküler modelleme aktivite çalışmaları sonuçlarının değerlendirilmesi, bu sayede obezite ile mücadele etmek için diyet ve egzersize ek olarak GOAT inhibitörlerinin kullanılması ve kilo kontrolü sağlanması amaçlanmıştır. Obezite tedavisi için önemli bir hedef olan ghrelin, kısa süreli olarak iştahı baskılamakta ve kilo kazanımını önlemektedir. Diğer obezite ilaçlarında görülen merkezi sinir sistemi ile ilgili yan etkileri önlemek için, doğrudan ghrelin reseptörü yerine ghrelini aktif hale getiren enzim hedef alınmıştır. Piyasada ghrelin tabanlı bir obezite ilacının bulunmaması bu hormon üzerinde odaklanmamızı sağlamıştır. Tez kapsamında GOAT enzimini inhibe edebilmek için 8 kimyasal molekül laboratuvar ortamında sentezlenmiş, moleküllerin analizi ve karakterizasyonu LC-MS ve NMR ile yapılıp yapıları aydınlatılmıştır. Sentezlenen bu moleküllerin GOAT reseptörüne bağlanma afinitesi moleküler modelleme yöntemleriyle test edilmiştir. Moleküler modelleme sonuçlarına göre laboratuvarımızda sentezlediğimiz ŞRPT-3 kodlu molekül GOAT inhibisyonu gösterebilecek özgün bir ilaç adayı olabileceği tespit edilmiştir. Obesity is a chronic disease that causes hyperlipidemia, Type-2 DM, hypertension and coronary heart diseases and the number of patients in the world and in our country has been increasing rapidly in recent years. Obesity brings with it economic problems. For all these reasons, researchers have focused on pharmacotherapy in obesity. Currently, there are obesity medicines named Orlistat, Lorcaserin, Phentermine/Topiramate, approved by the American Food and Drug Administration. However, these drugs have been reported to have cardiovascular and gastrointestinal side effects. In this study, the effect of GOAT (Ghrelin O-Acyl Transferase) enzyme on obesity was investigated. It is aimed to synthesize chemical molecules that will inhibit GOAT enzyme, to evaluate the results of molecular modeling activity studies in the computer environment, to use GOAT inhibitors in addition to diet and exercise to control obesity and to provide weight control. As an important target for the treatment of obesity, ghrelin suppresses appetite and prevents weight gain for a short time. In order to prevent central nervous system side effects seen in other obesity drugs, the enzyme activating ghrelin was targeted directly instead of ghrelin receptor. The lack of a ghrelin-based obesity drug on the market has enabled us to focus on this hormone. In order to inhibit the GOAT enzyme, 8 chemical molecules were synthesized in the laboratory. Analysis and characterization of the molecules were carried out by LC-MS and NMR. The binding affinity of these synthesized molecules to GOAT receptor was tested by molecular modeling methods. According to the results of molecular modeling, it is determined that the SRPT-3 encoded molecule synthesized in our laboratory may be a unique drug candidate that can show GOAT inhibition.
