Urfa peynirlerinden izole edilen staphylococcus aureus suşlarında enterotoksin üretim potansiyeli ve metisilin dirençliliği

Çalışma kapsamında, İstanbul ve Şanlıurfa'daki halk pazarı ve marketlerde satışa sunulan Urfa peyniri örneklerinden izole edilen Staphylococcus aureus suşları koagulaz ile DNaz enzim aktiviteleri ve metisilin dirençlilik özellikleri yönüyle değerlendirilmiştir. Peynir örneklerinden egg-yolk tellurit içeren Baird Parker Agar besiyeri kullanılarak izole edilen S. aureus suşlarına tavşan kan plazması kullanılarak koagulaz testi, DNaz agar ve Orsab agar kullanılarak ise sırasıyla DNaz aktivitesi ve metisilin dirençlilik testi yapılmıştır. Çalışma kapsamında analiz edilen toplam 52 adet Urfa peyniri örneğinin 48'inde (%92) ortalama 4.48±1.76 logkob/g düzeyinde S. aureus tespit edilmiş ve örneklerin sadece %25'inin Türk Gıda Kodeksi'ne uygun olduğu belirlenmiştir. Elde edilen 64 izolatın 22'sinin (%34.4) koagülaz pozitif, 31'inin (%48.4) DNaz pozitif olduğu, 20'sinin (%31.25) ise metisiline karşı dirençli olduğu tespit edilmiştir. Çalışma sonucunda analiz edilen Urfa peyniri örneklerinde enterotoksin üretme potansiyeli yüksek ve metisiline dirençli S. aureus izolatlarının bulunduğu, bu nedenle bu peynirlerin tüketiminin halk sağlığı için riskli olabileceği görülmüştür. In this study, the coagulase and DNase enzyme activities and methicillin resistance characteristics of S. aureus strains isolated from Urfa cheese samples sold in Istanbul and Şanlıurfa were determined. S. aureus strains isolated from cheese samples by using Baird Parker Agar containing egg yolk tellurite were tested for coagulase activity using rabbit blood plasma, DNase activity and methicillin resistance using DNase and Orsab agars. A total of 52 Urfa cheese samples were analyzed. Mean S. aureus load was 4.48±1.76 log cfu/g in 48 (92%) samples and only 25% of the samples were in good accordance with the limit imposed by the Turkish Food Codex. Of the 64 S. aureus isolates, 22 (34.4%) were coagulase positive, 31 (48.4%) were DNase positive, and 20 (31.25%) were resistant to methicillin. Results indicated that Urfa cheese samples may have high enterotoxin producing potential and methicillin resistant S. aureus isolates, so consumption of these cheeses could threat public health.
