dc.description.abstractObezite, ülkemizde ve dünyada artış göstermeye devam eden, bulaşıcı olmayan kronik bir hastalıktır. Çalışmamızda, kurkumin ve poliaminlerin obezite üzerine etkileri ve kan ve doku parametrelerine yansımalarının incelenmesi amaçlanmıştır. Çalışmada 28 adet Sprague-Dawley erkek sıçan deneysel obezite geliştirmek amacıyla 10 hafta yüksek yağlı diyet (%60) ile beslenmiş ve sonrasında kontrol (KONT); kurkumin (KUR); poliamin (POL); kurkumin ve poliamin (KUR+POL) grupları olmak üzere rastgele 4 gruba ayrılmıştır. Deney süresince tüm gruplar yüksek yağlı diyet ile beslenmiş ve takviyeler 6 hafta boyunca, 3 gün/hafta oral gavaj yolu ile uygulanmıştır. Sıçanlardan deney başlangıcında, obezite geliştikten sonra ve deney sonunda olmak üzere 3 kez kan alınmıştır. Karaciğer, böbrek ve yağ doku örneklerinde histopatolojik ve immünohistokimyasal incelemeler yapılmıştır. Sıçanlarda en düşük yem tüketimi KUR+POL grubunda (p<0,05); en düşük ağırlık POL grubunda, sonra sırasıyla KUR+POL, KONT ve KUR gruplarında gözlenmiş ancak gruplar arasında anlamlılık bulunmamıştır. Serum toplam poliamin düzeyleri ile obezite gelişimi arasında anlamlı bir ilişki tespit edilmemiş, N-asetil putresin (p<0,05) ve Gama Aminobutirik Asit (GABA) (p<0,001) düzeylerindeki artış ise anlamlı bulunmuştur. Gruplar arasında glukoz değerlerindeki artış anlamlı değildir. POL grubunda serum toplam kolesterol değeri KONT ve KUR+POL gruplarına göre anlamlı düzeyde (p<0,05) düşük bulunmuştur. Tüm gruplarda serum ALT değerleri başlangıca göre artmış olmakla birlikte gruplar arasında anlamlılık gözlenmemiştir. Yağ, karaciğer ve böbrek dokularında toplam histopatolojik skor KONT grubunda diğer gruplara göre anlamlı düzeyde artmış olup patolojik bulguların dejenerasyon yönünde olduğu saptanmıştır. Dokulardaki hasar KUR+POL grubunda diğer gruplara oranla daha fazla iyileşme yönünde ve Nükleer Faktör Kappa B (NF-κB) ekspresyonu anlamlı olarak düşük bulunmuştur (p<0,001). Sonuç olarak, POL takviyesinin toplam kolesterol düzeylerini azaltma; KUR+POL takviyelerinin birlikte kullanımının ise besin tüketimini azaltma ve inflamasyondan koruyucu etkileri görülmüştür. Kurkumin ve poliaminlerin obeziteye olan etkilerinin daha iyi anlaşılabilmesi için kapsamlı çalışmalara ihtiyaç duyulmaktadır.en_US
dc.description.abstractObesity is a non-communicable chronic disease that continues to increase in our country and around the world. In our study, it was aimed to investigate the effects of curcumin and polyamines on obesity and the reflection of these effects on blood and tissue parameters. 28 male Sprague-Dawley rats were fed high-fat diet (60%) for 10 weeks to develop experimental obesity and then they were randomly divided into 4 groups as the control group (CONT); curcumin (CUR); polyamine (POL); curcumin and polyamine (CUR+POL). During the experiment, all groups were fed with a highfat diet, supplements were administered by oral gavage for 3 days a week for 6 weeks. Blood was taken from rats at the beginning of the experiment, after the development of obesity, and at the end of the experiment, a total of 3 times. Histopathological and immunohistochemical examinations were performed on the liver, kidney, and adipose tissue. The lowest feed consumption in rats was recorded in the CUR+POL group (p<0.05); the group with the lowest weight after supplements was the POL group, then the CUR+POL, CONT, and CUR groups, respectively, but there was no significant difference between the groups (p>0.05). There was no significant relationship between serum total polyamine levels and the development of obesity (p>0.05), the increase in N-acetyl putrescine (p<0.05) and Gamma Aminobutyric Acid (GABA) (p<0.001) levels was found to be significant. The increase in glucose levels between groups was not significant. Total cholesterol value in the POL group was found to be significantly lower (p<0.05) compared to the CONT group and the CUR+POL group. Although serum ALT values increased in all groups compared to the baseline, no significance was observed between the groups (p>0.05). The total histopathological score in fat, liver and kidney tissue increased significantly in the CONT group compared to the other groups and the pathological findings were found to be in the direction of degeneration. In the CUR+POL group, damage to the tissues was in the direction of recovery compared to the other groups and the expression of Nuclear Factor Kappa B (NF-κB) was significantly low (p<0.001). As a result, it has been observed that POL supplementation reduced total cholesterol levels and the combined use of CUR+POL supplements may have protective effects on reducing food consumption and inflammation. Comprehensive studies are needed to better understand the effects of curcumin and polyamines on obesity.en_US
