dc.description.abstractBuğday gluteni sindirilebilir olması ve bağırsak immünitesinin modülasyonunda rol oynadığı bilinen amino asitlerin (glutamin, glutamik asit) kaynağı olması nedeniyle birçok memeli için iyi bir diyet protein kaynağı olarak kabul edilmektedir. Gluten tüketimi bugüne kadar birçok farklı rahatsızlık ile ilişkilendirilmiştir. Bunlardan en yaygın olanı ve en bilineni çölyak hastalığıdır. Fakat günümüzde, gluten tüketimi çölyak hastalığı dışında da bazı nörolojik rahatsızlıklar, depresyon, otizm, obezite gibi farklı sağlık problemleri ile ilişkilendirilmektedir. Glutensiz diyetin yaygınlığı artmakta, son on yılda, Amerika Birleşik Devletleri'nde neredeyse her üç kişiden biri, gluteni diyetlerinden çıkarmaya çalışmaktadır. Glutensiz diyetlerin kilo kontrolünde yaygın kullanımına rağmen, gluten ile obezite arasındaki ilişki hala tutarsızdır ve literatürde çok az kontrollü çalışma vardır. Bu çalışma yüksek yağlı diyet ile beslenen farelerde gluten tüketiminin vücut ağırlığı ve mikrobiyotaya etkisini saptamak amacıyla yapılmıştır. Çalışmada, 25 adet BALB/c fare kullanılmıştır. Fareler rastgele standart diyetin verildiği kontrol grubu (Grup 1), standart diyet ile 5 mg/gün glutenin verildiği (Grup 2), yüksek yağlı diyetin verildiği (Grup 3), yüksek yağlı diyet ile 5 mg/gün glutenin verildiği (Grup 4) olmak üzere 4 gruba ayrılmıştır. Her bir deneğin canlı ağırlığı haftalık periotlarda ölçülerek kayıt altına alınmıştır. Mikrobiyota analizleri için, her bir kafes geceden değiştirilerek haftada bir kez kafeslerden dışkı toplanmıştır. Mikrobiyota analizleri 16s rRNA yöntemi kullanılarak analiz edilmiştir. Çalışmada gluten ve yüksek yağlı diyet uygulamalarının kontrol grubuna göre istatistiki açıdan anlamlı derecede kilo kazanımına neden olduğu saptanmıştır (p<005). Toplam mezofilik aerobik bakteri sayısı parametresi açısından gruplar arasında istatistik açıdan anlamlı bir fark saptanmamıştır (p>005). Lactobacillus bulgaricus parametresi için diyetlere gluten eklenmesinin söz konusu mikrobiyolojik parametre ile pozitif yönde (gluten etkenin üremesini stimüle etmektedir) istatistik açıdan anlamlı olacak şekilde etkili olduğu elde edilmiştir (p<005). Yüksek yağlı diyet uygulamasının, bağırsakta bulunan toplam koliform grubu bakterilerinin üremesini engelleyici özellikte olduğu saptanmıştır (p<005). Gluten içeren diyet uygulamalarının, bağırsaktaki toplam koliform grubu bakteri üremesi üzerine negatif anlamda etkili olduğu bulunmuştur (p<005). Yüksek yağ ile diyet uygulamasının bağırsakta bulunan Esherichia coli üremesini engelleyici özellikte bir etkiye sahip olduğu elde edilmiştir (p<005). Gluten içeren diyet uygulamalarının, bağırsaktaki toplam Esherichia coli sayısı parametresinin üremesi üzerine negatif anlamda etkili olduğu görülmüştür (p<005). Glutensiz diyetin kullanıldığı durumlarda, mikrobiyotaya pozitif etkisi bulunan probiyotiklerin ve prebiyotiklerin beslenmeye eklenmesi önerilebilir.en_US
dc.description.abstractWheat gluten is considered a good dietary protein source for many mammals, as it is digestible and a source of amino acids (glutamine, glutamic acid) known to play a role in the modulation of intestinal immunity. Gluten consumption has been associated with many different ailments to date. The most common and well-known of these is celiac disease. However, today, gluten consumption is associated with different health problems such as some neurological disorders, depression, autism and obesity besides celiac disease. The prevalence of the gluten-free diet is increasing, with nearly one in three people in the United States trying to eliminate gluten from their diet in the last decade. Despite the widespread use of gluten-free diets in weight control, the relationship between gluten and obesity is still inconsistent, and there are few controlled studies in the literature. This study was conducted to determine the effect of gluten consumption on body weight and microbiota in mice fed a high-fat diet. In the study, 25 BALB/c mouse strains were used. The mice were randomly given the standard diet (Group 1), 5 mg/day gluten was given with a standard diet (Group 2), high-fat diet (Group 3), 5 mg/day gluten was given with a high fat diet (Group 4) divided into 4 groups. The body weight of each subject was measured and recorded in weekly periods. For microbiota analysis, each cage was changed overnight and feces were collected from cages once a week. Microbiota analyzes were analyzed using the 16s rRNA method. In the study, it was determined that gluten and high fat diet applications caused statistically significant weight gain compared to the control group (p <005). There was no statistically significant difference between the groups in terms of total mesophilic aerobic bacteria count parameter (p> 005). For the Lactobacillus bulgaricus parameter, adding gluten to the diets was found to be statistically significant in a positive way (in the direction of stimulating the growth of the agent by gluten) with the microbiological parameter in question (p <005). It has been determined that diet with high fat prevents the growth of bacteria from the total number of coliform groups in the intestine (p <005). It has been found that diet applications containing gluten have a negative effect on the growth of the parameter of total coliform group bacteria in the intestine (p <005). It has been found that diet with high fat has an effect preventing the growth of Esherichia coli in the intestine (p <005). It has been observed that diet practices containing gluten have a negative effect on the growth of the parameter of total Esherichia coli in the intestine (p <005). In cases where a gluten-free diet is used, it may be recommended to add probiotics and prebiotics that have a positive effect on the microbiota.en_US
