dc.description.abstractGünümüzde obeziteyle savaşmak için hem geleneksel hem de tıbbi temele dayalı çeşitli yöntemler kullanılmaktadır. Bu tezin amacı Türkiye'de geleneksel olarak zayıflama amacıyla tüketilen biberiye, papatya, mısır püskülü, maydanoz ve ceviz yaprağı bitki özütlerinin yağlı diyetle beslenen deneysel yolla obezleştirilmiş sıçanlarda kan şekeri, kan lipit profili ve vücut ağırlığına etkisinin incelenmesidir. Çalışma kapsamında 6-8 haftalık 30 adet Spraque Dawley dişi sıçan kullanılmıştır. Sıçanlardan 5 tanesi randomize olarak seçilerek kontrol grubu oluşturulmuş ve deney sonuna kadar standart pellet yemle beslenmiştir. Kalan 25 sıçan 4 hafta boyunca 25 g tereyağı/100 g yem olmak üzere yüksek yağlı diyetle beslenmişlerdir. Bu sürenin sonunda hayvanlar randomize olarak 6 gruba ayrılmıştır. Birinci grup yağlı beslenme kontrolü olarak ayrıldıktan sonra 2. grup biberiye, 3. grup papatya, 4. grup ceviz yaprağı, 5. grup maydanoz ve 6. grup mısır püskülü bitkilerinin özütleriyle 6 hafta boyunca gavaj yoluyla beslenmişlerdir. Hayvanların ağırlıkları her hafta başı tartılarak kaydedilmiştir. Çalışmanın başı, ortası ve sonunda kan örnekleri alınarak, serumlarında lipit profillerine ve kan glukoz değerlerine bakılmıştır. Bulunan veriler SPSS 18.0 programı ile analiz edilmiş ve istatistiksel olarak değerlendirilmiştir. Yağlı beslenerek obezleştirilen sıçanlarda vücut ağırlığı, kontrol grubu sıçanlarına oranla anlamlı artış göstermiştir. Kan değerlerinde obezleştirme süreciyle beraber total kolesterol ve HDL' de artış gözlemlenmiş; bitki özütlerinin diyetlerine eklenmesinden sonra papatya, maydanoz ve mısır püskülünün kan glukozunu anlamlı şekilde düşürdüğü görülmüştür. Karaciğer ağırlıkları arasındaki fark tüm gruplarda istatistiksel olarak anlamlı bulunmuştur. Yağlı beslenme ile obezleştirlen sıçanların diyetlerine eklenen papatya ve biberiye özütleri vücut ağırlıklarında azalmaya, papatya, mısır püskülü ve maydanoz özütleri verilen sıçanlarda kan glukozudan düşüşe neden olmuştur. Karaciğer ağırlıkları papatya ve biberiye özütü verilen sıçanlarda azalma gösterirken; ceviz yaprağı, maydanoz ve mısır püskülü özütleri verilenlerde arttığı gözlemlenmiştir. Kan lipit parametrelerinde bitki özütleri verilen gruplarda kontrol grubuna oranla herhangi anlamlı bir farklılık tespit edilmemiştir. Bu bitkilerin lipit metabolizması ve obezite üzerine etkilerinin incelenmesi için daha ileri çalışmalara ihtiyaç duyulmaktadır.en_US
dc.description.abstractIn our day, various methods are used traditionally and medically for the purpose of struggling with obesity. The purpose of this thesis is to analyze the effect of the plants of rosemary, chamomile, corn tassel, parsley and walnut leaf, which are consumed traditionally with the purpose of slimming in Turkey, on blood glucose, blood lipid profile and bodyweight in rats fed by fatty diet. Within the scope of this study, 30 female rats, which are 6-8 weeks old, from the type of Sprague-Dawley were used. By selecting five of the rats randomly, a control group was formed and the rats were fed by standard pellet feed until the end of the experiment. Other twenty-five rats were fed by highly fatty diet as 25 g butter/100 g feed during four weeks. In the end of this period, the animals were separated into six groups, randomly. After separating the first group as fatty diet control; the second group was fed by rosemary, the third group was fed by chamomile, the fourth group was fed by walnut leaf, the fifth group was fed by parsley and the sixth group was fed by corn tassel as extract and through enteral feeding during six weeks. Weights of the animals were calculated at the beginning of every week. By taking blood in beginning, middle and end of the study, the levels of blood glucose and the lipid profiles in the serum were analyzed. The results obtained were analyzed by SPSS 18.0 (a software package used for statistical analysis) and evaluated statistically. In terms of bodyweight, a significant difference was observed between the control group and the fatty-diet group. When all groups were compared, the difference in the weights of liver is statistically significant. In parallel with the period of occurring obesity, an increase has been observed in total cholesterol and HDL within the blood values; and it has been detected that chamomile, parsley and corn tassel decreased significantly the blood glucose after adding the extracts of the plants. In obesity, the effect of fatty diet and lipid metabolism are important issues. In our study, a significant difference has been observed in weight gain of the rats because of the fatty diet. By adding the extracts of before mentioned plants after the occurrence of obesity, any significant change has not been found in the bodyweights. In parallel with the period of occurring obesity, an increase has been observed in total cholesterol and HDL within the blood values; and it has been detected that chamomile, parsley and corn tassel decreased significantly the blood glucose after adding the extracts of the plants. Further studies are necessary for the purpose of analyzing the effects of these plants.en_US
